ROBO1

ROBO1全稱為Roundabout同源受體1，也被稱為DUTT2或SAX3，屬于軸突導向受體家族。ROBO1蛋白通過結合Slit配體調控神經細胞遷移和軸突導向，在胚胎發育中參與神經網絡構建，在腫瘤血管生成和癌細胞轉移中同樣發揮重要作用。ROBO1/Slit信號異常與肺癌、乳腺癌和結直腸癌等惡性腫瘤的侵襲性增強相關，其過表達可能通過激活MAPK和PI3K-AKT通路促進腫瘤細胞存活與遷移。ROBO1基因缺失或突變還與先天性心臟病、智力發育障礙存在關聯。目前針對ROBO1的藥物開發聚焦于抗腫瘤領域，已有研究團隊開發出特異性單克隆抗體，在動物模型中顯示出抑制腫瘤轉移的效果。ROBO1的雙重作用機制使其在神經修復和抗腫瘤領域都具有潛在開發價值。

IHC image of CSB-RA896760MA1HU diluted at 1:30 and staining in paraffin-embedded human brain tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-Mouse IgG labeled by HRP and visualized using 0.05% DAB.

Overlay Peak curve showing Hela cells surface stained with CSB-RA896760MA1HU (red line) at 1:50. Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1μg/1*10⁶cells) for 45 min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1μg/1*10⁶cells) used under the same conditions. Acquisition of >10,025 events was performed.
